Our Technology
From affinity maturation to protein engineering, variants will always be needed to find the required activity. Ninthbio's Homology Path DNA synthesis tools design your DNA populations to be assembled from the least number of oligos possible. Our approach is so aggressive, it typically saves 90% when compared to synthesis by a service provider. We design the oligos to assemble with a simple PCR reaction; any molecular biology lab can take control of their DNA synthesis pipeline.

Homology Path Software
Homology Path designs the oligos needed to synthesize a population of DNA sequences by finding the shared scaffolds and designing the build around those common areas.
1
Import FASTA File
Import the sequences of interest as a single FASTA file.
2
Check Complexity
Our complexity analysis shows you where the potential sequence issues are, so you can optimize to best meet your synthesis and expression requirements
3
Generate Population Design
Run the oligo designer, it will automatically find the possible recycling and build the most economical design
4
Oligo Order Is Generated
The list of oligos needed are generated in a format that can be submitted directly to your oligo vendor for production
5
Pool the Oligos
Pooling instructions are generated for you to pool the oligos by hand or by automation; we accommodate all automation formats
6
Run PCR
Run standard PCR reactions on the oligo pools just as you would a DNA template
DNA Assembly
Our pooling instructions and assembly are robust and ideal for automation platforms. Our customers have used almost every imaginable transfer method, from pipetting for small projects, or using the Labcyte Echo for pooling thousands of reactions per day.
Oligo Pooling
The instructions generated from Homology Path can be uploaded directly to most automation systems for the pooling of your oligos.
PCR Assembly
The oligos pools are designed to assemble in a high fidelity robust reaction. This assembly reaction is perfectly suited for automated Thermal Cyclers; both stand-alone, or integrated with your automation.
